Protein antibiotics are species-specific antimicrobials deployed by Gram-negative bacteria to kill their neighbours during competition for resources. My laboratory is exploring the import mechanism of nuclease colicins, protein antibiotics that specifically target nucleic acids in the cytoplasm of Escherichia coli. As well as trying to understand how these folded, 60-kDa proteins translocate into bacteria we are also exploiting colicins as molecular probes of fundamental cell envelope processes. My talk will focus on the latter area and in particular some recent work where we have discovered how Gram-negative bacteria organise their outer membrane proteins.
